Biological Dose Assessment by Chemical Induction of Premature Chromosome Condensation

Authors

Abstract:

Dose assessment of radiation victims is a key element in medical management of radiation accidents. At high radiation doses (> 6 Gy), dicentric assay lose its efficiency and premature chromosome condensation (PCC) assay was proposed to overcome the restrictions of dicentric assay. It was shown that PCC ring is more suitable and simpler than dicentric for biodosimetry in high radiation doses. In this study, we established standard dose response curve using chemical PCC ring assay on human peripheral blood lymphocytes for X-ray in the dose-interval of 0–10 Gy. The chemical PCC assay performed on Peripheral blood lymphocyte of four healthy donors after irradiation with different doses of X-rays (0-10 Gy) and the IAEA recommendations for production of an in vitro dose response curve were followed. The dose response curve was fitted based on the linear quadratic model and practicable for high radiation doses. Chemical PCC ring assay is applicable with no need to any special equipment even after supra lethal radiation doses.

Construction of a dose – response curve by induction of premature chromosome condensation for biological dosimetry

Premature Chromosome Condensation (PCC) appears to have a possible utility for biological dosimetry purposes. The PCC technique may be adapted for cases of suspicion of overexposure where sampling is performed at least one day after an accident. For this purpose, human blood samples were exposed in vitro to 60Co up to 10 Gy and the PCC technique was performed immediately after irradiation. Anal...

Induction of premature chromosome condensation (PCC) by calyculin A for biodosimetry

Background: Premature chromosome condensation (PCC) is a method for interphase chromosome analysis in biodosimetry. This study was performed to verify the usefulness of PCC induced by calyculin A in human peripheral blood lymphocytes (pbl) for biological dosimetry and possible construction of dose-response curve. Materials and Methods: Peripheral blood was obtained from a healthy donor and expo...

Construction of Calibration Curve for Premature Chromosome Condensation Assay for Dose Assessment

Cytogenetic dosimetry plays an important role in the triage and medical management of affected people in radiological incidents/accidents. Cytogenetic biodosimetry uses different methods to estimate the absorbed dose in the exposed individuals, and each approach has its advantages and disadvantages. Premature chromosome condensation (PCC) assay presents several advantages that hopefully fulfill...

Induction of Premature Chromosome Condensation (PCC) by Calyculin A for biodosimetry

B iological dosimetry of ionizing radiation is generally performed by scoring dicentrics and/or translocations in chromosome from peripheral blood lymphocytes (pbl) at the first mitosis following in vitro growth stimulation. It is well known that after irradiation, in this procedure mitotic index is low in some individuals, so the scored mitoses might not be representative of the exposed cell p...
